Thiophilic adsorption chromatography on Fractogel EMD TA

چکیده
Introduction High performance affinity chromatography is one of the most powerful techniques for the isolation of biomolecules. The high efficiency of the method is based on the specific interaction of a covalently bound ligand with its counterpart immobilized on a chromatographic support. This adsorption is due to electrostatic forces between charged groups, hydrogen bonds and nonpolar or hydrophobic interactions. Apart from biospecific ligands such as receptors, substrates, cofactors and antibodies there is a wide range of applications using pseudo biospecific compounds like dyes, metal ions, amino acids or sulphur containing groups as ligands. The latter one is designed especially for the efficient isolation of antibodies. One important advantage of these pseudo biospecific ligands is their chemical stability.

Removal of Aggregates of Monoclonal Antibodies by Ion Exchange Chromatography
Abstract—The primary objective of this work was to study the effect of resin chemistry, pH and molarity of binding and elution buffer on aggregate removal using Cation Exchange Chromatography and find the optimum conditions which can give efficient aggregate removal with minimum loss of yield. Four different resins were used for carrying out the experiments: Fractogel EMD SO3(S), Fractogel EMD ...
متن کاملInexpensive synthetic-based matrix for both conventional and rapid purification of protein A- and tandem affinity purification-tagged proteins
Immunoglobulin G (IgG)-Sepharose is often used for purification of protein A- and tandem affinity purification (TAP)-tagged proteins from eukaryotic cells, but because it is based on an agarose matrix, it is not always optimal for all proteins. Synthetic matrices such as IgG-Dynabeads have improved properties over IgG-Sepharose but are generally expensive. Here we describe the preparation and p...
متن کاملImproved performance of micro-fabricated preconcentrators using silica nanoparticles as a surface template.
A new approach of enhancing the adsorption capability of the widely used polymer adsorbent Tenax TA poly(2,6-diphenylene oxide) through its deposition on a nano-structured template is reported. The modified Tenax TA-coated silica nanoparticles (SNP) are incorporated as an adsorbent bed in silicon based micro-thermal preconcentrator (μTPC) chips with an array of square microposts embedded inside...
متن کاملComparison of Adsorption/Desorption of Volatile Organic Compounds (VOCs) on Electrospun Nanofibers with Tenax TA for Potential Application in Sampling
The objective of this study was to compare the adsorption/desorption of target compounds on homemade electrospun nanofibers, polystyrene (PS) nanofibers, acrylic resin (AR) nanofibers and PS-AR composite nanofibers with Tenax TA. Ten volatile organic compounds (VOCs) were analyzed by preconcentration onto different sorbents followed by desorption (thermal and solvent orderly) and analysis by ca...
